This enzyme catalyzes the GalNAc-transfer from UDP-GalNAc to Ser or Thr residue in the protein. The product is GalNAc-alpha-O-Ser/Thr structure, so-called Tn-antigen, which is the base structure of mucin type O-glycan. The acceptor substrate (protein) is typically Mucin. GALNT6 is the sixth member of this enzyme family. This was discovered as a highly homologous gene with GALNT3, but its function is not redundant with T3.

UDP-GalNAc:polypeptide N-acetylgalactosaminyltransferase
J. Biol. Chem. 1999-09-01 ;274(36):25362-70.
Cloning and characterization of a close homologue of human UDP-N-acetyl-alpha-D-galactosamine:Polypeptide N-acetylgalactosaminyltransferase-T3, designated GalNAc-T6. Evidence for genetic but not functional redundancy.
