β1,4-N-acetylgalactosaminyltransferase-III (B4GALNT3) transfers GalNAc from UDP-GalNAc to GlcNAcβ-R, synthesizing N,N'-diacetyllactosamine, GalNAcβ1→4GlcNAc (LacdiNAc) on N- and O-glycans. B4GALNT3 is also known as B4GalNac-T3. LacdiNAc structures are also synthesized by its homologue B4GALNT4, and occur on various glycoproteins including lutropin (LH), thyrotropin (TSH), zona pellucida 3 (ZP3), and extracellular matrix glycoproteins. LDN can be further modified with sulfation, sialylation, and fucosylation, indicating important roles of LacdiNAc structures in vivo.
