Initially, 3GnT1 (iGnT; B3GNT1) was isolated by the expression cloning method, and it was reported as an enzyme that transfers GlcNAc to the N-acetyllactosamine structure ([LacNAc: Galβ1-4GlcNAc]), and by further extending, biosynthesizes the poly-N-acetyllactosamine (polylactosamine, [Galβ1-4GlcNAc]n), the structure of which is the repeated structure of the N-acetyllactosamine. However, as it was later reported by “in vitro” experiments, this enzyme is the B4GAT1 enzyme involved in the synthesis of the α-dystroglycan by the transfer a glucuronic acid by the b1-4 linkage. The B4GAT1 enzyme is involved in the synthesis of the structure of GlcA ([GlcAb1-4Xyla1-R), which is the first starting point of the repeating structure of the disaccharide ([GlcAb1-4Xylb1-3]n) synthesized by the LARGE/LARGE2.
