MFNG encodes Manic Fringe; O-fucosylpeptide 3-beta-N-acetylglucosaminyltransferase. Fringe (in mammals, there are Lunatic Fringe, Manic Fringe, and Radical Fringe) is a β1,3-N-acetylglucosaminyltransferase involved in the synthesis of O-linked fucose glycans in the EGF domain of Notch protein, and was discovered as a gene that causes a mutation in Drosophila melanogaster that results in abnormal dorsal-ventral boundary formation during feather formation. Fringe transfers GlcNAc toward O-fucose synthesized by POFUT1, are essential for the activation of Notch signalings through the ligands such as Delta and Serrate/Jagged.
